(PH098) Immunoassay (ELISA) for the coplanar Polychlorinated Biphenyls (PCBs).

ABSTRACT- The development of a highly sensitive enzyme-linked immunosorbent assay (ELISA) based on polyclonal antibodies for coplanar PCB congeners is described. The ELISA utilizes antibodies developed in rabbits using a PCB 169-protein immunogen in combination with a PCB 77-HRP enzyme conjugate. The ELISA was tested for its cross-reactivity profile using 20 selected congeners. The assay was highly specific for non-ortho substituted (coplanar) congeners and did not recognize the more abundant but less toxic non-coplanar PCB congeners or organochlorine compounds. The sensitivity of the ELISA for PCB 169 and PCB 126 is in the low parts per trillion. Performance of the assay and its validation will be presented.
