(PH145) Profiling of the tiger worm (Eisenia fetida) proteins in relation to organic waste toxicity.

ABSTRACT- The hazardous effect of organic wastes-contaminated soil on terrestrial organisms was assessed. Four types of municipal sewage sludge (MSS), industrial sewage sludge (ISS), leather processing sludge (LPS), and alcohol fermentation processing sludge (AFPS) along with the standard pig manure compost (PMC) were applied and mixed with soil (15cm in depth). The appropriate amount of waste as 12.5, 25 and 50 tons of dry matter were treated annually from 1994 to 2001. Heavy metal contents, residual organic compounds, and earthworm numbers were determined. ISS, LPS, and AFPS significantly affected the growth of earthworms. However, in the first 4 years, earthworms in PMC and MSS were grown well and the numbers increased. Therefore, the earthworms grown in the two wastes such as PMC and MSS were investigated through proteomic analysis. Their different protein expressions were assessed using a nanospray ionization tandem mass spectrometry (MS/MS) coupled with nanoscale two-dimensional HPLC. As a result, more than 300 proteins were identified from the approximately 600 peptides assigned in the study. In the whole extract of the earthworm grown in MSS contaminated soil, acetylcholine receptor, divalent cation transporter, GABA receptor and membrane calcium ATPase decreased. However, enzymes involved in the gluconeogenesis were increased. In addition, acetylcholinesterase, carboxylesterase, and carbonic anhydrase were also induced. The similar results may be obtained from yeast proteome with starvation stress. This expression profile of E. fetida will serve as the valuable data of a proteome expressed in the earthworm.
