(PT015) The effect of chlorophenols toward cellular growth of Bacillus subtilis subsp. subtilis.

ABSTRACT- It is well known that halogenated aromatic compound like o -chlorophenol is one of the environmental pollutants. The nineteen kinds of chlorophenols as a precursor of dioxin compounds and phenol as the basic compound were used to the cellular growth test of Bacillus subtilis subsp. Subtilis (NBRC3007). And comparison of these antibacterial concentrations to the physical properties of these compounds was done. Finaly we try to estimate the antibacterial concentrations from that physical property. In this study, 1x104 of vegetative cells were cultured with 0.0001% to 2% of those chlorophenols or not. After 24hr at 37°C, the numbers of vegetative cells were counted using CFU method. As the basically, obtained antibacterial concentrations of phenol showed 0.080%(about 800ppm). Three monochlorophenols showed the range of 0.030% to 0.050%. Six dichlorophenols showed the range of 0.0030% to 0.040%. Six trichlorophenols showed the range of 0.00040% to 0.0180%. Three tetrachlorophenols showed the range of 0.00045% to 0.020%. Pentachlorophenol showed 0.00070%. 3,4,5-Trichlorophenol showed most strong effect by the concentration of only 0.00040%. And 2,3,4,5-tetrachlorophenol showed strong effect by the concentration of 0.00045%. The antibacterial concentrations of those chlorophenols were different by the position and the number of chlorine to the phenol. The physical properties of these compounds were listed as mp(°C), bp(°C), solubility to the water(mg/ml), pKa and the partition coefficients of n-Octanol/water(Pow). Some compound showed good correlation between the antibacterial concentration and solubility to the water. Low solubility compound showed strong effect to vegetative cells. The antibacterial concentrations of these compounds were strongly correlated to the partition coefficients of Pow. The compound showed the high value of Pow had strong effect to vegetative cells. It is possible to estimate the antibacterial concentrations of chlorophenols by the solubility to the water and the partition coefficients of n-Octanol/water.
