(M/FF243) Investigation of the mutagenicity of water in top floor water boxes.

ABSTRACT- Following upon the increase of high floor building and the deve1opment of industries, more and more populations will drink the water in top f1oor water boxes. Thus the peop1es pay attention to the problems of pollution from secondary provided water. We had studied the mutagenicity of the top floor box water on five high floor buildings in Hangzhou, so as to protect the hea1th of the residents in city. We used the micronucleus test of root tips of vicia faba, which was 1ow cost, sensitive, convenient, standard. The tested water samples were prepared by the method of concentration with XAD--8 nonpolarzation resin. In study, According to Chen G. R.(1987), seeds of bean vicia faba germinated in water, Cultured in 500 ml beaker containing running water at 25-C.;When the secondary roots were 2--3cm long, The roots were expousured in the tested water for 4--6hr, The plants repaired in the fresh water for 24hr. Then the root tips (about 1cm long) were fixed, softened, stained (Feulgen) and pressed slices (conventional method) throughout the study.3 root tips were used in every sample, The micronucleus frequency of every root tip were studied in 1000 cells of root tips. The resu1ts were analysed by F test. This study showed that the micronucleus frequency of the water of the top f1oor water boxes from Dongmao questhouse and Zhi-- jiang hotel were 3.48% and 1.92% respectively. Compared with the individual negative control, the difference was significant (p<0. 01). The results of this study suggested that the provided water of above two hotels were polluted by the variant mutagens to a different degree. This study provided an important scientific basis for prevented the secondary pollution of provided water and reduced the morbidity of cancers.
