(40-04) Determination of polyether antibiotics and tiamulin from manure and soil samples.

ABSTRACT- Antibiotics such as polyethers (salinomycin and monensin) and pleuromutilins such as tiamulin are used to treat infectious diseases in livestock. Some are used growth promoters (salinomycin) as well. Both groups of compounds are heavily used in the feeding phase of pigs, and to less extend in cattle. The respective animals absorb most of the compounds quickly but they do excrete the compound after some days or weeks with excretion rates of 50-90% of the applied amount. The respective antibiotics are trapped in the manure, some undergo some microbial metabolism and are brought with the manure to the fields. This study focuses on the determination of these synthetic antibiotics in the agricultural environment. A typical pig farm in Germany grows about 800 pigs each 9 months. Depended on the infections in the livestock about 1-5 kg active ingredients are used per compound during the feeding phase. In this study methods we present methods based on ASE and HPLC-MS/MS which have been elaborated to analyse polyether antibiotics as well as tiamulin in soil. Liquid partitioning together with an SPE-clean up and HPLC-APCI-MS/MS detection was used for the extraction of manure. The respecticve recovery rates varied from 60-90%. The clean up was optimised for the compounds in question, as well as the respective MS/MS transitions. Optimisation of MS transitions for the quantification of the poyether antibiotics as well as Tiamulin gave the following conditions: Compound (transition, collision energy) Salinomycin (768.7->733.6 amu, -22eV) Monensin (688.5->461.3 amu,-27eV) Tiamulin (494.6->192.3 amu,-27eV)
