ABSTRACT- A Detailed Review Paper (DRP) for an avian two-generation reproductive/developmental toxicity test with endocrine endpoints is being developed by the U.S. Environmental Protection Agency. The avian DRP will summarize, explain, and document relevant principles, methods, and techniques recommended for development of an initial protocol(s), and will identify those issues that may require prevalidation studies to adequately address. Emphasis is placed on the relevance and sensitivity of the candidate assays/endpoints for identifying chemicals that affect endocrine systems in bobwhite quail and Japanese quail. The DRP will provide a basis for selecting the most promising methods for assessing chemical effects on avian endocrine systems and for determining future studies required to resolve outstanding issues relative to method development, including the need for additional targeted prevalidation studies. This information will be used to support the design of a draft protocol for use in the validation phase of protocol design, with the ultimate goal of developing future EPA/OECD test method guidelines. The preliminary findings from initial DRP efforts will be presented.
