STUDY ON BCL-2 GENE EXPRESSION IN APOPTOTIC HUMAN THECAL CELLS INDUCED BY TRANSFORMING GROWTH FACTOR-1 PLUS TRANSFORMING GROWTH FACTOR- IN VITRO.

Objective: To investigate the apoptosis of human thecal cells induced by trasforming growth factor-1 (TGF-1) plus trasforming growth factor-(TGF-) in vitro and bcl-2 gene expression in apoptotic human thecal cells.

Methods: Human thecal cells were harvested by enzymatic digestion from human follicles of healthy fertile women,which were obtained in laparotomy. The apoptosis of human thecal cells was induced by TGF-1 plus TGF- in vitro culture and was tested by lightmicroscopy, electromicroscopy and apagargel electrophoresis. Expression of bcl-2 gene in apoptotic human thecal cells was observed by means of immunocytochemical image analysis and morphological measurement.

Results:Combined treatment with TGF-1 plus TGF- cause dramatic morphological and biochemical chatacterics of human apoptotic thecal cells. Bcl-2 gene was expressed in cytoplasm of normal human thecal cells. Treatment with TGF-1 plus TGF- lead to a decline in expression of bcl-2 in cytoplasm but a positive staining in the nucleus whereas nuclei of control groups did not have positive staining of bcl-2.

Conclusions: It was suggested that TGF-1 and TGF- could induce the apoptosis of human thecal cells in vitro. The induction of apoptosis of human thecal cells may be regulated not only by decline of bcl-2 expression, but may also involve its redistribution from the cytoplasm into the nucleus.
