Abstract: Our objective was to determine whether physiological doses of E2 and P4 affect nuclear and cytoplasmic maturation of rhesus monkey oocytes in vitro. All immature oocytes were collected from untreated non-cycling adult and prepubertal rhesus monkeys during the summer and the follicles (1000 m or greater) were retrieved from excised ovaries. In experiment 1, 357 cumulus-oocyte complexes (COCs) from adult monkeys were randomly cultured in: 1) control, mCMRL-1066 + FSH + LH; 2) mCMRL-1066 + FSH + LH + P4; 3) mCMRL-1066 + FSH + LH + E2; 4) CMRL-1066 + FSH + LH + P4 + E2, and 5) CMRL-1066 + P4 + E2. In experiment 2, 161 COCs from prepubertal monkeys were matured in: 1) control, mCMRL-1066 + FSH + LH; 2) mCMRL-1066 + FSH + LH + P4, and 3) mCMRL-1066 + FSH + LH + E2. The concentrations of FSH, LH, P4 and E2 were 5 g/ml, 10 g/ml, 3 g/ml and 100 ng/ml, respectively. In experiment 1, no difference was found in the rates of oocyte meiotic maturation, fertilization and embryo development to 16-cells among the five groups (P>0.05). However, embryo development beyond the morula stage was significantly different (P<0.05) between group 1 and groups 2-5 (morula + blastocyst: 21.4%, 44.4%, 32.4%, 42.5% and 34.8%, respectively;), although not different among groups 2-5 (P>0.05). In experiment 2, oocytes matured with E2 showed the highest rate (P<0.05) of development to at least 9-cell embryos (32.1%, 37.5% and 56%, in groups 1-3, respectively) and to morula and blastocyst (10.7%, 20.8%, and 40%, respectively) after IVF. Oocytes matured with P4 had more potential to develop into morulae compared with control (20.8% versus 10.7%; P<0.05). These results indicate that 1) P4 and E2 can significantly increase cytoplasmic maturation (developmental competence) of non-stimulated rhesus monkey oocytes; 2) COCs from adult and prepubertal rhesus monkeys have different sensitivities to stimulation by E2 and P4. .
