THE EFFECTS OF THE CATALASE AND XANTHINE ON IN VITRO CULTURE OF PORCINE FOLLICULAR OOCYTES.

Abstract:
The objective of the present study were to examine the relationship between catalase and xanthine on in-vitro maturation of porcine oocytes. The follicular oocytes were cultured in TC-199 medium with catalase (0.7mg/ml), xanthine (5 mM) or catalase plus xanthine. At 48 h after culture, the proportions of oocytes matured to metaphase-II stage were significantly (P<0.05) higher in medium with control group (72%), catalase (73%) or catalase+xanthine (70%) than in oocytes cultured with xanthine (54%). The effect of catalase on morphological changes during in-vitro culture with various periods in porcine oocytes were examined. At 72 h after culture, the maturation rates increased in medium with and without catalase. However, degenerated oocytes were increased with culture periods prolonged, the proportion was was significantly (P<0.05) higher in medium without (47%) than with (28%) catalase at 120 h after culture. At 96 and 120 h after culture, parthenogenetic oocytes were observed, and the proportions were higher in medium with (24 and 35%) than without (13 and 22%) catalase. In another experiment, when oocytes were cultured in medium with xanthine, the rates of degenerated oocytes were increased with culture periods, and were higher in medium without than with catalase. At 72 h of culture, parthenogenetic oocytes were observed and increased with culture periods prolonged, but there were no significantly different between with and without catalase. These results indicate that porcine oocytes may respond to maturation stimulus by 72 h of culture, and oocyte degeneration were inhibited by catalase in medium with xanthine. Parthenogenetic oocytes can be obtained with high rates from oocytes cultured in medium with catalase.
