(M523) PROTEIN PROFILES OF OVARIAN FOLLICULAR FLUID DURING DIFFERENT PHASES OF ESTROUS CYCLE IN MURRAH BUFFALOES.

ABSTRACT- The present study was designed to compare the protein concentrations and protein profiles of the follicular fluid collected from ovaries of the different phases of the estrous cycle and also from true acyclic ovaries of Murrah buffaloes. Ovaries (n = 300) were collected from non-pregnant Murrah buffaloes soon after slaughter and their ultrasonography was performed. Ovaries were classified to be in the follicular, early luteal, mid luteal, late luteal phases and true acyclic phase. Follicular fluid was aspirated from follicles and protein concentration was measured. Blood samples were collected during estrous phase of buffaloes (n= 5) and serum was separated. SDS-PAGE using Laemmli discontinuous buffers system was performed for resolving protein in the samples of both follicular fluid and blood serum. Follicular fluid protein concentrations was found to be significantly (P < 0.05) lower in acyclic ovaries than normally cycling ones. SDS-PAGE of the follicular fluid of each phase revealed 17 protein bands in the molecular size range of approximately 15 kDa to 145 kDa. Two bands of approximately 107 kDa and 37 kDa size present only in follicular fluid and two bands of approximately 38 kDa and 35 kDa size only in serum samples. These findings suggested that (a) follicular fluid protein concentration did not differ significantly during different phases of estrous cycle, but was significantly lower (P < 0.05) in acyclic ovaries than that of cyclic ones and (b) there might be expression of ovary specific proteins locally during different phases of estrous cycle in Murrah buffaloes.
